Abstract:

رفتار هسته در تلیوسپور، بازیدیوسپور و میسلیوم­های Ustilago maydis مورد بررسی قرار گرفت. جهت کشت تلیوسپورها از محیط PDA + 10% دکستروز استفاده گردید. مطالعه رفتار هسته با استفاده از میکروسکوپ اپی­فلورسنت و رنگ­آمیزی با دو رنگ اتیدیوم برماید و اکردین اورنج صورت پذیرفت. هسته در تلیوسپور کروی و بزرگ و اکثر فضای داخلی آن را اشغال نموده است. بازیدیوسپورها (اسپوریدیوم) به صورت غیرانگلی و به روش جوانه­زنی رشد می­کنند و بازیدیوسپورهای جوانه­زده با زاویه º35 الی º45 از اسپوریدیوم مادری قرار دارند. هسته در بازیدیوسپور بیضوی شکل و در هنگام تشکیل اسپوریدیوم جدید، به محل تشکیل اسپوریدیوم مهاجرت و پس از تقسیم میتوز یک هسته به درون اسپوریدیوم در حال تشکیل منتقل می­شود. ریسه­ها درون بافت آلوده به دو صورت مشاهده شدند. برخی دو­هسته­ای و ظریف و به اشکال متفاوت شاخه­های متورم درون سلولی تا زوائد انگشت مانند و برخی ریسه­ها در محل تشکیل تلیوسپور تک­هسته­ای و ضخیم و دیپلوئید بودند.ریسه­های U. maydis حاصل از کشت بافت آلوده در محیط کشت عموماً دو­هسته­ای و هسته­ها در ارتباط نزدیک با یکدیگر و در نزدیکی دیواره سلولی قرار داشتند. درون حفره­های مملو از تلیوسپورهای جوان و بالغ، همه ریسه­ها در حال تبدیل شدن به تلیوسپور بودند و ریسه دو هسته­ای مشاهده نگردید.
